Kajian Diversitas Keratinase Berdasarkan Urutan Residu Asam Amino dan Struktur Protein

Abstrak: Miotermus taiwanensis memiliki aktifitas keratinase sangat tinggi dan menarik untuk digunakan sebagai landasan kajian diversitas
keratinase. Penelitian ini melaporkan analisis global keratinase berdasarkan urutan residu asam amino dan struktur tiga dimensinya.
Sekuen protein diunduh dari pangkalan data NCBI (Center for Biotechnology Information) dengan menggunakan urutan protein
keratinase dari M. taiwanensis. Seratus protein diseleksi dengan identitas minimal 65 % dan analisis urutan residu asam amino
menunjukkan pemisahan protein tersebut menjadi dua kelompok, sedangkan analisis domain mengelompokkan semuanya ke dalam
protease serin. Analisis kesamaan struktur 3D hanya memiliki kesamaan di bawah 40 % dan keunikan domain membedakannya dari protease lainnya.

Abstract : Miotermus taiwanensis has a very high keratinase activity and it is interesting to use as a foundation for keratinase diversity
studies. This study reported a global keratinase analysis based on the sequence of amino acid residues and their 3D structure. The protein
sequence was downloaded from the NCBI (Center for Biotechnology Information) database using the protein sequence of keratinase from M. taiwanensis. One hundred proteins were selected with a minimum identity of 65 % and the analysis of the amino acid sequence showed the separation of the protein into two groups, whereas the domain
analysis grouped them all into serina proteases. The similarity analysis of 3D structure had only similarity below 40 % and doman uniqueness of its structure that distinguish it from other proteases.

Strategi Analogi dalam Pembelajaran Praktikum Sel Volta

Abstrak: Strategi analogi di dalam pembelajaran praktikum dapat dimanfaatkan untuk memperjelas fenomena makroskopis. Setengah sel
oksidasi dan setengah sel reduksi pada rangkaian sel volta menghasilkan voltase, hanya dipahami sebagai perubahan energi kimia menjadi energi
listrik. Strategi analogi dapat mengekplisitkan fenomena mikroskopis dan simbolik melalui penyajian konsep analog yang familiar dan
memiliki kemiripan dengan konsep target. Konsep analog berupa aliran air terjun atau aliran dana antara bank dan nasabah peminjam dapat
dipergunakan untuk menjelaskan konsep target aliran elektron. Penyajian deskripsi persamaan dan perbedaan antara kedua konsep
(target dan analog) diperlukan untuk meminimalisir terjadinya kesalahan pemahaman konsep. Strategi analogi diharapkan mempermudah peserta didik untuk mengkonstruksi dan mencapai pemahaman konsep.

Abstract: The purpose of using analogy in laboratory practice was to explain macroscopic phenomenon. Reduction and oxidation half-cells in
voltaic cell produce voltage which was just understood by students as a change from chemical energy to electrical energy. Analogy strategy
could make microscopic and symbolic phenomenon easier for students to understand by presenting analog concepts which was more familiar
for students and which had similarity with target concepts. The chosen analog concepts were waterfall flow or finance flow of a bank which can
be used to explain electron flow as target concept. Explanation of similarity and difference of target and analog concept which were needed to minimize misconception. Analogy strategy could be expected help students construct the concept and reach conceptual understanding.
